II. Biological Activity & Target
Biological Activity: Pemetrexed Disodium is an antifolate agent that exerts antitumor effects by inhibiting key enzymes in the folate metabolism pathway.
Target: It primarily acts on thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT), with Ki values of 1.3 nM, 7.2 nM, and 65 nM for inhibiting these enzymes, respectively.
III. Pharmacological Effects
Pemetrexed Disodium is a novel and classic antifolate drug with antitumor activity that may be due to the simultaneous and multiple inhibition of several key folate-requiring enzymes by its polyglutamated metabolites.
In vivo, the polyglutamation and polyglutamated metabolites of Pemetrexed Disodium may play an important role in determining its selectivity and antitumor activity.
