  2. Mechanism of Action:

    • Gemcitabine is a deoxycytidine analog that is activated within cells by nucleoside kinases to form gemcitabine diphosphate (dFdCDP) and gemcitabine triphosphate (dFdCTP).

    • dFdCDP inhibits ribonucleotide reductase, thereby inhibiting DNA synthesis.

    • dFdCTP is incorporated into replicating DNA chains, terminating chain elongation and inducing cell apoptosis.
