Pharmacological Mechanism

  1. H1 Receptor Antagonism: Azelastine competitively and selectively binds to H1 receptors, blocking the actions of histamine, a key mediator of allergic reactions.

  2. Mast Cell Stabilization: It enhances the stability of mast cell membranes, inhibiting the release of histamine, leukotrienes, and other inflammatory mediators.

  3. Antagonism of Other Inflammatory Mediators: Azelastine significantly inhibits the actions of bradykinin, substance P, serotonin, and platelet-activating factor.

  4. Suppression of Airway Inflammatory Cells: It significantly reduces eosinophil infiltration in animal models of asthma.

  5. β2-Adrenergic Receptor Sensitization: Enhances the sensitivity of β2-adrenergic receptors in the airways, reversing downregulation caused by chronic β2-agonist use.

Indications
