Pharmacological Action
Propylthiouracil inhibits the thyroid peroxidase system, blocking the iodination of tyrosine and the condensation of iodinated tyrosines within the thyroid gland, thus suppressing the synthesis of thyroid hormones. Additionally, at high doses (up to 700 mg daily), it can reduce the conversion of T4 to T3 in peripheral tissues. It also exhibits immunosuppressive effects. Since Propylthiouracil only inhibits the synthesis of thyroid hormones and has no effect on pre-existing hormones, it may take 3-4 weeks to observe therapeutic effects.
Indications
Propylthiouracil is suitable for various types of hyperthyroidism, particularly in the following situations:
Mild to moderate cases with small to moderate thyroid enlargement.
Adolescents, children, and the elderly.
Recurrence after thyroid surgery, where radioactive iodine-131 (¹³¹I) treatment is not suitable.
