Mechanism of Action: Raltitrexed is a folate-based thymidylate synthase (TS) inhibitor that undergoes rapid and extensive metabolism in vivo to form more active polyglutamated derivatives. It also inhibits dihydrofolate reductase (DHFR). As a potent antimetabolite antitumor agent, raltitrexed selectively inhibits thymidylate synthase, thereby inhibiting DNA and RNA synthesis, demonstrating antitumor activity in vivo.
