Pharmacological Action
Iguratimod functions as an anti-rheumatic agent primarily through the following mechanisms:
Inhibition of COX-2 Activity: Iguratimod effectively inhibits COX-2 activity with an IC50 value of 20 μM (7.7 μg/mL), while showing no effect on COX-1. This allows it to reduce inflammation and pain without the common gastrointestinal side effects associated with non-steroidal anti-inflammatory drugs (NSAIDs).
Inhibition of MIF (Macrophage Migration Inhibitory Factor): Iguratimod is also an inhibitor of MIF with an IC50 value of 6.81 μM. By inhibiting MIF, it further mitigates inflammatory responses.
