Uses and Efficacy
Gliquidone belongs to the second-generation sulfonylureas class of oral hypoglycemic drugs, along with glibenclamide, gliclazide, and glipizide.
It is indicated for the management of non-insulin-dependent (Type 2) diabetes mellitus, particularly suitable for patients with short disease duration, mild conditions, elderly individuals over 60 years old, those with poor constitution, malnutrition, and those accompanied by significant cardiovascular and cerebrovascular atherosclerosis.
For Type 2 diabetic patients with mild to moderate renal impairment, gliquidone is a preferred option due to its primarily hepatic metabolism and minimal (5%) renal excretion.
Mechanism of Action
Gliquidone stimulates insulin secretion from pancreatic β-cells and enhances insulin sensitivity in peripheral tissues.
It potentiates insulin receptor function and insulin action post-receptor.
Early in treatment, it primarily promotes endogenous insulin secretion; later on, it primarily improves insulin sensitivity in surrounding tissues.
