Pharmacological Action
Gliclazide belongs to the sulfonylurea class of oral antidiabetic drugs. Its main pharmacological actions include:
Hypoglycemic Effect: It stimulates the secretion of insulin from the pancreatic beta cells, inhibits hepatic glycogenolysis and gluconeogenesis, thereby reducing both fasting and postprandial blood glucose levels. This effect is dependent on the residual ability of the pancreatic beta cells to synthesize and secrete insulin.
Vasculoprotective Effect: It reduces platelet adhesion and aggregation, decreases thromboxane levels, enhances endothelial cell plasminogen activator activity, leading to increased fibrinolytic capacity, slows down microvascular endothelial cell fibrosis, and improves endothelial function.
