Pharmacological Action:
Doxofylline, a derivative of methylxanthine, acts primarily as a bronchodilator. It relaxes smooth muscles of the bronchus by inhibiting phosphodiesterase and adenosine A1 receptor, thereby exerting its bronchodilatory effect. Its bronchodilatory and smooth muscle relaxant actions are 10-15 times stronger than theophylline, with a prolonged duration of action. Additionally, Doxofylline possesses antitussive properties not found in theophylline and provides bronchial dilation without affecting adenosine's effects on the heart, thus avoiding extrapulmonary adverse reactions such as those observed with caffeine.
