Mechanism of Action:
Sofosbuvir is a prodrug that, once metabolized in the liver, is converted into the active antiviral agent, GS-461203 (also known as 2'-deoxy-2'-α-fluoro-β-C-methyluridine-5'-triphosphate).
This active metabolite acts as an inhibitor of the NS5B RNA-dependent RNA polymerase, which is essential for the viral replication of Hepatitis C.
By incorporating into the HCV RNA chain during replication, it causes chain termination, thus inhibiting the ability of the Hepatitis C virus to replicate its RNA.
Clinical Use:
Sofosbuvir is highly effective when used in combination with other antiviral medications such as ribavirin, pegylated interferon, and other direct-acting antivirals (DAAs) like ledipasvir.
It is used in various genotypes of Hepatitis C, often achieving high cure rates.
