Tigecycline is a derivative of minocycline, with a glycylamido substitution at the C-9 position of the minocycline structure. This substitution pattern is not found in any natural or semisynthetic tetracycline drug.
The presence of a glycylamido group enhances its antibacterial activity and reduces bacterial resistance.
Pharmacological Effects
Tigecycline exerts its antibacterial action by binding to the bacterial 30S ribosomal subunit, preventing the entry of aminoacyl-tRNA molecules into the A-site of the ribosome, thereby inhibiting bacterial protein synthesis.
Tigecycline has a broad spectrum of activity covering G+ cocci, G- bacilli, MRSA, ESBL-producing Enterobacteriaceae, Stenotrophomonas maltophilia, and multi-drug-resistant Acinetobacter baumannii.
